The advances in antibody engineering have now facilitated a more efficient and generally applicable method to produce Fv fragments. Fv fragment is the smallest unit of immunoglobulin molecule with function in antigen-binding activities. An antibody in ScFv format consists of variable regions of heavy (VH) and light (VL) chains, which are joined together by a flexible peptide linker that can be easily expressed in functional form in E. coli, allowing protein engineering to improve the properties of ScFv such as increase of affinity and alteration of specificity.

There are several advantages of phage-displayed ScFv over monoclonal antibodies.

  • Firstly, phages are more stable and can be stored up to several years at 4°C
  • Secondly, they can be produced rapidly and inexpensively just by infecting the E. coli
  • Thirdly, their genes can be easily manipulated and they can be produced by circumventing hybridomas and immunization
  • Also, higher affinity mutants of ScFv can be generated through site-directed mutagenesis which is much easier and simpler to be performed

Basically, scFv antibodies produced from phage display can be genetically fused to the marker proteins, such as fluorescent proteins or alkaline phosphatase. Alternatively, antibody fragments could also be applied in the construction of immunotoxins, therapeutic gene delivery, and anticancer intrabodies for therapeutic purpose.
