CAR-T cells have quickly shown great potential to manipulate the immune system and regulatory agencies are starting to approve CAR-T cell therapies to treat diseases such as large B-cell lymphoma. Moreover, different from CAR-T cells, CAR-NK cells can still exert their "natural" anti-leukemia effect in case the tumor antigen targeted by CAR is downregulated. However, understanding the variability of sample preparation and establishing quality measures that are predictive of potency, safety and consistency for manufacturing purposes are still challenging issues.

  • CAR Expression Detection

Evaluating CAR expression is an essential step in the production of CAR-T cells. This is often done by flow cytometry, using protein L, anti-Fab antibodies or target antigens as detection antibodies. Among these common choices, target antigens are widely considered to be the best option, because it offers high specificity and minimal background staining.

  • Cytokine Release Analysis

Cytokine Release Syndrome (CRS) is usually due to on-target effects induced by binding of the bispecific antibody or CAR-T cell receptor to its antigen and subsequent activation of bystander immune cells and non-immune cells, such as endothelial cells.

  • Cytotoxicity in vitro Assay

In this assay, long-term cytotoxic function and proliferative capacity of CAR-T cells is examined in vitro over 7 days with additional tumor targets administered to the co-culture every other day. This assay can be coupled with profiling T cell activation, exhaustion and memory phenotypes.
